A laboratory freeze dryer is easiest to operate when the run is treated as a sequence of checks. The exact screen names and limits depend on the supplied configuration, so the approved equipment manual always takes priority. Before loading the freeze dryer
Confirm that the chamber, drain and condenser are clean and dry. Inspect the door gasket, vacuum hose, pump oil level and electrical supply. Samples should already be frozen below their complete freezing point unless the system has controlled in-situ freezing.
Record the sample identity, container, fill depth and estimated water load. A condenser rated in kilograms per 24 hours is not permission to load that much ice at once; the instantaneous vapor load and available condenser surface also matter.
- Use compatible flasks, trays or partially stoppered vials.
- Keep fill depth consistent within a study.
- Do not process unknown solvents without a compatibility review.
Start cold, then establish vacuum
Start the refrigeration stage and allow the ice condenser to reach the run-specific acceptance temperature before opening the product path. After loading, seal the chamber and start the vacuum pump according to the machine sequence.
Watch both condenser temperature and chamber pressure. A pressure that stalls high can indicate warm product, a leak, wet pump oil, an open drain or excessive vapor generation. Do not compensate by immediately adding more heat.
Monitor primary and secondary drying
For shelf-controlled work, product temperature should remain below the formulation limit during primary drying. Compare product probes, shelf temperature and chamber pressure trends rather than relying on elapsed time alone.
Secondary drying usually uses a higher shelf temperature at low pressure to remove more strongly bound moisture. The endpoint and maximum product temperature must come from the product method, not from a universal recipe.
Unload, defrost and document
Backfill or vent the chamber using the specified clean gas or filtered air sequence. Remove the product promptly and close or package it before it can absorb room moisture.
Isolate the vacuum pump before defrost. Allow ice to melt through the designed drain; never chip the condenser coil. Clean spills, dry the chamber and record alarms, pressure behavior and any deviation before the next batch.
